ACID TOLERANCE

Acid resistance of Bifidobacterium breve HH-BR26 powder was carried out at either pH=2.0 and pH=3.0 for up to 4 hours. The microbial enumeration of Bifidobacterium breve HH-BR26 should be performed no more frequently than at the initial(as control), 1 hour, 2 hour, and 4 hour time points. The Fig 1 illustrates that Bifidobacterium breve HH-BR26 can survive at low pH conditions, which means this strain has good acid resistance.

BILE SALT TOLERANCE

Bile salt resistance of Bifidobacterium breve HH-BR26 powder was carried out at either 0.2% and 0.3% bile salt for up to 4 hours. The microbial enumeration of Bifidobacterium breve HH-BR26 should be performed no more frequently than at the initial(as control), 1 hour, 2 hour, and 4 hour time points. The Fig 2 illustrates that Bifidobacterium breve HH-BR26 can survive at high concentration of bile salt, which means this strain has good bile salt resistance.

STORAGE STABILITY

Bifidobacterium breve HH-BR26 powder were sealed in aluminum foil bag, maintained at -18℃ for 24 months for long-term stability test. The microbial enumeration of Bifidobacterium breve HH-BR26 should be performed no more frequently than at the initial, 1, 2, 3, 6, 9, 12, 15, 18, 21, 24 month time points. According to Fig 3, the potency of Bifidobacterium breve HH-BR26 powder was 2.57×10¹¹ CFU/g. After a storage period of 24 months, the potency of the powder was 1.38×10¹¹CFU/g. It means that this strain can remain stable for long time if stored at low temperature.

1. Antioxidant capacity

In healthy people, a balance exists between free radical production and antioxidant defenses. Disturbances in this balance may contribute to disease. Excessive oxygen exposure, physiologic stress, infection, and trauma are associated with ROS and RNS overproduction. Through colorimetric experiments (Table 1), we found that live B.breve HH-BR26 and its cell fragments showed strong DPPH scavenging activity amd can prevent lipid-peroxidation.
